Chromogenic complete ELISA-kit for determination ADAMTS-13 activity in human plasma.
Test principle
GST-vWF73 substrate is incubated in micro wells coated with monoclonal anti-GST-antibodies. After a washing step the diluted samples are added into the wells and the plate is incubated. ADAMTS-13 cleaves the GST-vWF73 substrate. Unbound substances are removed by washing. A peroxidase conjugated detecting antibody is added, which detects the cleavage site on GST-vWf73. After a second washing step, added enzyme substrate reacts with the conjugate. The intensity of the resulting colour is proportional to the activity of ADAMTS-13 in the sample.
