Emicizumab calibrator is a calibrator plasma for the calibration of a modified one-step FVIII activity assay for the determination of active emicizumab concentrations in citrated plasma from a clotting time measurement.

Test principle

Emicizumab calibrator enables the quantification of the degree of correction of the APTT-based one-step assay by emicizumab in the sample. The calibrator is diluted in different concentrations. Each concentration is combined and incubated with FVIII-deficient plasma and APTT reagents. The reaction is started by the addition of CaCl2. The clotting time is then measured. The correction level of APTT is related to the emicizumab activity and is converted to µg/ml using the calibration curve.
