Complete ELISA-kit for quantitative determination of the activity of human urokinase-type plasminogen activator (u-PA) as well as the total u-PA concentration in human plasma.

Test principle

The diluted samples are incubated in micro wells coated with monoclonal anti-u-PA-antibodies. After binding of u-PA to the capture antibody, unbound substances are removed by washing and a mix of Glu-plasminogen and plasmin-substrate is added. After a photometric activity determination the plate is washed, leaving the bound u-PA on the plate. A secondary, peroxidase conjugated, anti-u-PA-antibody is added which recognizes bound u-PA. After a washing step, added substrate reacts with the conjugate. The intensity of the resulting colour is proportional to the content of u-PA in the sample.
