Capture and detecting antibodies for the construction of an ELISA for the quantitative determination of free and total Protein S in citrated human plasma.
Test principle
Micro wells are coated with Protein S antibodies (capture antibody), to which the Protein S from the sample binds. Unbound substances are removed by washing. A peroxidase conjugated detecting antibody is added and binds to the Protein S/capture antibody complex. After a washing step, added enzyme substrate reacts with the conjugate. The intensity of the resulting colour is proportional to the Protein S content in the sample. Free Protein S can be determined, after the samples, controls and reference plasmas have been pre-treated with PEG.
