Capture and detecting antibodies for the construction of an ELISA for the quantitative determination of α₁-antitrypsin.
Test principle
Micro wells are coated with α₁-antitrypsin antibodies (capture antibody), to which the α₁-antitrypsin from the sample binds. Unbound substances are removed by washing. A peroxidase conjugated detecting antibody is added, which bind to the α₁-antitrypsin / capture antibody complex. After a washing step, added enzyme reacts with the conjugate. The intensity of the resulting colour is proportional to the α₁-antitrypsin content in the sample.
