Complete fluorogenic ELISA-kit for the determination of ADAMTS-13 activity and antigen concentration in human plasma.

Test principle

The undiluted samples are incubated in micro wells coated with monoclonal anti-ADAMTS-13-antibodies. After binding of ADAMTS-13 to the capture antibody, unbound substances are removed by washing. Added activity substrate reacts with the bound ADAMTS-13 and the resulting fluorescence is proportional to the ADAMTS-13 activity of the sample. After a washing step, a peroxidase conjugated detecting antibody is added and binds to the ADAMTS-13 /capture antibody complex. After a second washing step, added substrate reacts with the conjugate. The intensity of the resulting colour is proportional to the antigen content of ADAMTS-13 in the sample.
