Test principle
Urokinase-Plasminogen-Activator (u-PA) is incubated in micro wells coated with monoclonal anti-u-PA-antibodies. After binding of the u-PA to the capture antibodies unbound substances are removed by washing. Following this prediluted samples are added and incu-bated. Hereby u-PA/PCI-complexes are formed. After another washing step a monoclonal peroxidase-conjugated anti-PCI detecting antibody is added and binds to the complex bound PCI. After a washing step added enzyme substrate reacts with the conjugate. The intensity of the resulting colour is proportional to the Protein C Inhibitor content in the sample.
